Ocular histopathology in animals experimentally infected with Mycobacterium leprae and M. lepraemurium 1. Mycobacterium leprae and M. Iepraemurium infections in the mouse

چکیده
At varying periods of time following the successful establishment of systemic infections with Mycobacterium leprae or M. lepraemurium in the mouse and the nine-banded armadillo eyes were examined by light microscopy. Inoculation of bacilli was by the intravenous or intraperitoneal route or directly into the hind footpads; eyes were not directly inoculated in this study. During periods of up to 3 years under laboratory conditions no animal showed evidence of impaired vision or blindness, and the external appearance of both eyes was normal. The ocular histopathology and the sites of accumulation of bacilli are described. In immunologically normal mice infected with M. lepraemurium bacilli were much commoner in extraorbital tissues, but they were, nevertheless, found in various tissues within the orbit, including the ciliaiy body and sclera. In immunologically normal mice (and one rat) injected with M. leprae of human origin no bacilli were found in the eye, but in mice immunologically depressed by thymectomy and total body irradiation considerable numbers of bacilli were present in the iris and ciliary body and also in the limbal cornea. In the armadillo bacilli were found in large numbers in virtually all tissues except the lens, retina, optic nerve, and aqueous and vitreous humours, but the uveal tract was heavily involved. Findings are discussed in relation to the great frequency of ocular involvement and the importance of immune-complex disease in patients with lepromatous leprosy, and to factors which may favour the localisation and multiplication of Mycobacteriumn leprae in the eye.

Evaluation of the Immune Response in Mice Infected
Mycobacterium marinum is a photochromogenic myco bacterium capable of indu c in g a se lflimitin g loca li zed infection within the mouse foot pad ( 1.4 ). Because this infection is íwalogous to the M. leprae infection in the mouse, it has been suggested as an a lternative experimenta l model for studying the role of host immunity to M. leprae infection in the mouse foot pad (1 2. 16. 19). Numerous studies have so far been conducted on the cultivation of Mycobacterium leprae and Mycobacterium lepraemurium. However, no media-bound method of cultivation has been established. A number of investigators have reported that M. lepraemurium may be cultivated by cell culture technics. The method employed by Chang (1. 2. 3 ), Chang and Neikirk (4), Chang, Anderson and Vaituzis (~), in whic...

Inability to cultivate Mycobacterium leprae in vitro has been regarded as the bottleneck which obstructs research on modes of transmission, pathogenesis, immunization and the treatment of leprosy . The bottleneck in the cultivation problem always has been that of gaining significant information from an organism that has given no useful response to intensive inquiries by the disciplines of bioch...
متن کاملA RAPID AND SIMPLE METHOD OF DNA EXTRACTION FROM CLINICAL SPECIMENS CONTAINING MYCOBACTERIUM LEPRAE FOR PCR ASSAYS
We describe a rapid, simple, and reliable procedure for routine isolation of Mycobacterium leprae DNA from slit-skin swab specimens. This one-step DNA extraction method is based on the utilization of ChelexR 100, a chelating ion exchange resin. Slit-skin swab specimens from seven untreated leprosy patients at Baba Baghi Leprosy Hospital in Iran were processed by this procedure. The polymer...
